Rami Malek är född i maj 1981, och hans fyra minuter yngre tvillingbror lyssnar till namnet Sami. Rami är av egyptisk härkomst, men han är examinerad vid Univeristy of Evansville, Indiana. Han började sin filmkarriär med gästskådespeleri i några amerikanska serier, bland annat Gilmore Girls. Hans filmdebut kom år 2006 med rollen som Farao i Night at the museum.
